目的调查徐州地区汉族人群血小板特异性抗原(HPA)基因多态性及特点,为人类遗传学提供资料,为本地区临床血小板免疫性减少症患者提供血小板输注配型参考。方法采用序列特异性引物聚合酶链反应(PCR-SSP)对徐州地区100名无血缘关系的汉族成年人进行HPA1~17等位基因分型。在包含引物混合液的96孔反应板中,按照相同的循环条件扩增PCR产物。结果在徐州地区汉族人群的HPA1~17系统中,呈多态性分布的等位基因是HPA1a、HPA2a、HPA3a、HPA4a、HPA5a、HPA6a、HPA15a,其频率分别为0.995 0、0.935 0、0.590 0、0.9950、0.980 0、0.975 0、0.575 0,HPA7~14、16~17呈单线性分布。结论徐州地区汉族人群HPA的基因有地区特点,人类血小板抗原HPA的基因具有民族多态性。
Objective To investigate the polymorphisms and characteristics of platelet-specific antigen (HPA) gene in Han population in Xuzhou, and to provide information for human genetics and to provide a reference for platelet transfusion in patients with clinical platelet immunopathies in the region. Methods 100 unrelated Han adults in Xuzhou district were genotyped with HPA1-17 alleles by PCR-SSP. In a 96-well reaction plate containing primer mix, PCR products were amplified according to the same cycling conditions. Results The alleles of HPA1 to HPA1-17 distributed in Han population in Xuzhou were HPA1a, HPA2a, HPA3a, HPA4a, HPA5a, HPA6a and HPA15a with frequency of 0.995 0, 0.935 0 and 0.590 0, 0.9950,0.980 0,0.975 0,0.575 0 HPA7 ~ 14,16 ~ 17 showed a single linear distribution. Conclusion The genetic characteristics of HPA in Han population in Xuzhou area are regional and the genetic polymorphism of human platelet antigen HPA is ethnic.
