目的研究丙烯酰胺染毒对小鼠睾丸组织半胱氨酸天冬氨酸蛋白酶3(caspase-3)与角蛋白8(cytokeratin 8,CK8)表达的影响。方法将40只健康5~6周龄清洁级雄性昆明小鼠随机分为4组,分别为对照(蒸馏水)组和低(10 mg/kg)、中(20 mg/kg)、高(40 mg/kg)剂量丙烯酰胺染毒组,每组10只。采用腹腔注射方法进行染毒,染毒容量为0.01 ml/g,连续染毒7 d。采用免疫组织化学方法测定caspase-3和CK8在小鼠睾丸组织中的表达,并进行病理学观察。结果各丙烯酰胺染毒组出现不同程度的生精小管管腔变形,生精细胞排列紊乱等。与对照组比较,高剂量丙烯酰胺染毒组小鼠睾丸组织CK8表达水平明显下降,而caspase-3表达水平明显升高,差异均有统计学意义(P<0.01)。随着丙烯酰胺染毒浓度的升高,小鼠睾丸组织CK8表达水平呈下降趋势,caspase-3表达水平呈上升趋势。结论丙烯酰胺导致的小鼠睾丸组织细胞凋亡与caspase-3高表达,CK8低表达有关。
Objective To investigate the effect of acrylamide on the expression of cystatin 8 and cytokeratin 8 (CK8) in mouse testes. Methods Forty healthy male Kunming mice aged 5-6 weeks were randomly divided into 4 groups: control (distilled water), low (10 mg / kg), medium (20 mg / kg) / kg) dose of acrylamide exposure group, each group of 10. The method of intraperitoneal injection was used. The exposure volume was 0.01 ml / g and continuous exposure for 7 days. Immunohistochemistry was used to detect the expression of caspase-3 and CK8 in mouse testes and pathological observation. Results The acrylamide exposure group showed different degrees of spermatogenic small tube lumen deformation, spermatogenic cells arranged disorder. Compared with the control group, the expression of CK8 in the testis tissue of the high dose acrylamide group was significantly decreased, while the expression of caspase-3 was significantly increased, the difference was statistically significant (P <0.01). With the increase of acrylamide concentration, the expression level of CK8 in mouse testis decreased and the expression level of caspase-3 showed an upward trend. Conclusion Acrylamide-induced apoptosis in mouse testis tissue is related to the high expression of caspase-3 and the low expression of CK8.
