目的:通过观察苏木乙酸乙酯提取物对同种异位心脏移植急性排斥反应模型大鼠移植心肌ICAM-1 mRNA表达的影响,探讨其免疫抑制作用的机理,为苏木的新药研发提供重要的理论和实验依据。方法:以Wistar大鼠为供体,SD大鼠为受体,制备大鼠同种异位心脏移植模型,造模成功后,采用Real Time-PCR法测定ICAM-1 mRNA的表达。结果:与模型对照组相比,苏木组、Cs A组及苏木加半量Cs A组均能明显抑制ICAM-1 mRNA的表达,而苏木乙酸乙酯提取物及苏木乙酸乙酯提取物加半量Cs A抑制ICAM-1 mRNA表达的作用相当,且在抗排斥反应时,苏木乙酸乙酯提取物可以减少Cs A的用量。结论:在大鼠心脏移植模型中,苏木乙酸乙酯提取物能抑制同种异位心脏移植模型大鼠心肌ICAM-1 mRNA的表达,减轻移植心脏的急性排斥反应。
OBJECTIVE: To investigate the effect of ethyl acetate extract on the expression of ICAM-1 mRNA in the myocardium of allograft recipients with acute allograft heart transplantation and to investigate its mechanism of immunosuppression in order to provide an important Theoretical and experimental basis. METHODS: Wistar rats were used as donors, SD rats were used as recipients to prepare rat allograft heart transplantation model. After successful modeling, the expression of ICAM-1 mRNA was determined by Real Time-PCR. Results: Compared with the model control group, the expression of ICAM-1 mRNA was significantly inhibited in the Hematoxylin and CsA groups and in the CsA group and the semi-CsA groups, while the extracts of ethyl acetate and ethyl acetate The effect of adding CsA on the mRNA expression of ICAM-1 was almost the same, and ethyl acetate extract could reduce the amount of CsA during anti-rejection. Conclusion: In the rat heart transplantation model, ethyl acetate extract can inhibit the expression of ICAM-1 mRNA in the myocardium of allograft heart transplantation rats and relieve the acute rejection of the transplanted heart.
