目的分析汞作业工人周围血中T细胞受体-Vβ(TCR-Vβ)亚家族表达模式的变化,了解汞对T细胞免疫功能的影响。方法利用逆转录聚合酶链反应分析15例接汞工人周围血TCR-Vβ亚家族的分布情况,另选15名一般办公室健康人员作为对照组。结果 15例汞作业工人周围血单核细胞(PBMC)可检测到15~23个Vβ亚家族,中位数为21个亚家族,与对照组(可检测全部24个亚家族)比较,差异有统计学意义(P=0.000)。TCR-Vβ亚家族在汞接触工人中分布具有比较集中的趋势,全部样本均未能检测到Vβ6亚家族的表达,而Vβ16表达频率低,仅为20%,Vβ12表达频率为33%,而Vβ4的表达频率则为40%。结论本研究提供了汞接触对周围血T细胞Vβ亚家族出现选择性表达的现象,这可能是汞对T细胞损伤的一些早期表现。
Objective To analyze the expression of TCR-Vβ subfamily in peripheral blood of mercury exposed workers, and to understand the effect of mercury on T cell immune function. Methods The distribution of TCR-Vβ subfamily in 15 cases of mercury-exposed workers was analyzed by reverse transcription-polymerase chain reaction (RT-PCR). Fifteen general office health workers were selected as the control group. Results 15 to 23 Vβ subfamilies were detected in peripheral blood mononuclear cells (PBMCs) from 15 cases of mercury workers, with a median of 21 subfamilies. Compared with the control group (all 24 subfamilies were detectable), the difference was Statistical significance (P = 0.000). The distribution of TCR-Vβ subfamily in mercury exposure workers was concentrated. All samples failed to detect the expression of Vβ6 subfamily, while the expression of Vβ16 was low at only 20%. The frequency of Vβ12 expression was 33% The frequency of expression was 40%. Conclusions This study provides a demonstration of the selective expression of the Vβ subfamily of peripheral blood T lymphocytes by mercury exposure, which may be some of the early signs of mercury damage to T cells.
