目的探讨姜黄素对急性坏死性胰腺炎小鼠的保护作用及其机制。方法随机将40只健康的KM小鼠分为对照组、急性坏死性胰腺炎(ANP)模型组和低、高剂量姜黄素处理组各10只。在造模成功后12 h采集血样并处死小鼠,取胰腺组织观察病理损伤情况并进行病理学评分,分别采用全自动生化分析仪、ELISA双夹心技术、RT-PCR法、Western blot法检测血清淀粉酶活性、血浆TNF-α水平、各组胰腺组织NF-κB mRNA及NF-κB p65总蛋白和胞核蛋白的表达。结果血清淀粉酶、TNF-α水平,胰腺组织病理学评分及NF-κB表达ANP组较对照组明显增高(P<0.05),低、高剂量姜黄素治疗组较ANP组显著降低(P<0.05)。结论姜黄素可有效减轻急性坏死性胰腺炎小鼠胰腺损伤,其机制可能与降低NF-κB活性有关。
Objective To investigate the protective effect of curcumin on acute necrotizing pancreatitis in mice and its mechanism. Methods Forty healthy KM mice were randomly divided into control group, acute necrotizing pancreatitis (ANP) model group and low and high dose curcumin treated groups. Blood samples were collected 12 h after model establishment and sacrificed. The pancreatic tissue was taken to observe pathological lesions and pathological scores. Serum was detected by automatic biochemical analyzer, ELISA double sandwich technique, RT-PCR, and Western blot. Amylase activity, plasma TNF-α level, expression of NF-κB mRNA, total NF-κB p65 protein and nuclear protein in pancreatic tissue of each group. Results Serum amylase, TNF-α levels, pancreatic histopathological scores and expression of NF-κB in ANP group were significantly higher than those in control group (P<0.05). Low and high dose curcumin groups were significantly lower than those in ANP group (P<0.05). ). Conclusion Curcumin can effectively reduce pancreatic injury in mice with acute necrotizing pancreatitis, and its mechanism may be related to the decrease of NF-κB activity.
