目的:研究甘草中治疗湿热痹症的有效成分—三萜皂苷类成分的提取纯化工艺。方法:采用正交试验法,分别对溶媒量、提取时间、提取次数进行考察,采用大孔树脂吸附技术进行纯化,并用高效液相色谱法进行含量分析。结果:甘草治疗湿热痹症的有效成分的最佳提取工艺是采用50%乙醇溶液,用10倍量溶剂回流水浴提取3次,每次2h;最佳纯化工艺是采用X-5树脂吸附,取浓度为0.2g.mL-1溶液上样,最大上样量为50mL,后用50%的乙醇5倍量洗脱。结论:最佳条件下,甘草中治疗湿热痹症的有效成分能被充分提取纯化。
Objective: To study the extraction and purification of triterpene saponin, the active ingredient in licorice for the treatment of diarrhea. Methods: Orthogonal test was used to investigate the amount of solvent, the extraction time and the number of extraction. The macroporous resin was used for purification and the content was analyzed by high performance liquid chromatography. Results: The optimal extraction process of licorice root active ingredients was 50% ethanol solution, 10 times the amount of solvent reflux water bath extraction 3 times, each 2h; the best purification process is the use of X-5 resin adsorption, the concentration 0.2g.mL-1 solution loading, the maximum sample volume of 50mL, followed by 50% ethanol 5 times the amount of elution. CONCLUSION: Under the optimal conditions, the active ingredients of licorice for treating diarrhea can be fully extracted and purified.
