目的:建立反相HPLC法测定人血浆中利福喷丁的浓度。方法:以利福平为内标,采用ZORBAXSB-C18色谱柱(4.6mm×250mm,5μm),以甲醇-0.02mol·L-1磷酸缓冲液(67:33,v/v;pH7.0)为流动相,流速1.0ml·min-1,柱温40℃,340nm处紫外检测。结果:日内平均回收率为97.5%,RSD=4.1%(n=5),日间平均回收率为95.6%,RSD=5.4%(n=5),线性范围为0.54-17.28μg·ml-1(r=0.9995)。结论:本法简便快速,灵敏度高,重现性好,适用于利福喷丁化疗方案的临床监测及其药动学研究。
Objective: To establish a reverse phase HPLC method for the determination of rifapentine in human plasma. Methods: Rifampicin was used as an internal standard and was eluted with methanol-0.02 mol·L-1 phosphate buffer (67:33, v / v, pH7.0) on a ZORBAXSB-C18 column (4.6 mm × 250 mm, As mobile phase, flow rate 1.0ml · min-1, column temperature 40 ℃, UV detection at 340nm. Results: The average daily recovery was 97.5%, RSD = 4.1% (n = 5), day average recovery was 95.6%, RSD was 5.4% (n = 5) and the linear range was 0.54-17.28μg · ml-1 (r = 0.9995). Conclusion: This method is simple, rapid, sensitive and reproducible. It is suitable for the clinical monitoring and pharmacokinetic study of rifapentine chemotherapy.
