目的 :探讨红藻氨酸 (KA)致痈大鼠海马神经细胞凋亡与野生型p5 3(wtp5 3)蛋白表达的关系。方法 :采用KA诱导癫痫发作大鼠模型。以TUNEL方法标记DNA片段 ,原位检测凋亡细胞 ;免疫组化SP法检测wtp5 3蛋白。结果 :注射KA 72h后 ,癫痫发作大鼠海马CA1区凋亡神经细胞达高峰及wtp5 3蛋白表达在 2 4h大量出现 ;wtp5 3蛋白表达与细胞凋亡呈正相关。结论 :癫痫神经细胞凋亡与wtp5 3表达密切相关 ;痫性发作使wtp5 3表达上调 ,从而诱导神经细胞凋亡。
Objective: To investigate the relationship between the apoptosis of hippocampal neurons and the expression of wild type p5 3 (wtp5 3) protein in kainic acid (KA) -stimulated rats. Methods: A rat model of seizure was induced by KA. The DNA fragments were labeled by TUNEL method and apoptotic cells were detected in situ. The wtp5 3 protein was detected by immunohistochemical SP method. RESULTS: After 72 h KA injection, the number of apoptotic neurons reached peak in hippocampal CA1 area and the number of wtp5 3 protein appeared in 24 h in seizure rats. The expression of wtp5 3 protein was positively correlated with apoptosis. CONCLUSION: The apoptosis of epileptic nerve cells is closely related to the expression of wtp5 3. Epileptic seizure up-regulates the expression of wtp5 3 and induces neuronal apoptosis.
