以披碱草×野大麦杂种F1幼穗为外植体,设置不同的激素浓度,获得适宜的诱导培养基,并选用长度不同的幼穗为外植体,确定出较为合适的外植体长度。愈伤组织经过多次继代后,细胞生长缓慢,通过更换培养基使其恢复生长。实验结果表明,最适培养条件为:以长为2-3cm幼穗为外植体,MS+3mg/L2,4-D为诱导培养基。MS+2,4-D 3mg/L+KT 0.5mg/L+CH 300mg/L与MS+2,4-D 4mg/L均可改善经过长期继代生长质量较差的愈伤组织的生长情况。
Using the young spikelets of Elymus dahuricus × Hordeum brevisubulatum as explants, different hormone concentrations were set up to obtain suitable induction medium, and the young spikelings with different length were used as explants to determine the appropriate explant length . After multiple subcultures of callus, the cells grew slowly and regained their growth by changing the medium. The experimental results showed that the optimal culture conditions were as follows: Young spikes of 2-3 cm were used as explants and MS + 3 mg / L 2,4-D as the induction medium. MS + 2,4-D 3mg / L + KT 0.5mg / L + CH 300mg / L and MS + 2,4-D 4mg / L all improved the growth of callus .
