[目的]分析血管紧张素Ⅱ受体拮抗剂厄贝沙坦(Irbesartan)对X射线照射大鼠空肠上皮细胞(IEC-6)诱导纤溶酶原抑制物-1(PAI-1)、白介素-6(IL-6)mRNA表达和IEC-6增殖活性的影响。[方法]用RT-PCR证实IEC-6表达血管紧张素转换酶1受体(AT1受体)mRNA和AT2受体mRNA。用RT-PCR观察10Gy X射线照射IEC-6细胞前后PAI-1、IL-6mRNA的表达情况,同时观察照射+厄贝沙坦对IEC-6细胞PAI-1、IL-6mRNA表达的影响。采用MTT法观察对细胞增殖活性的影响。[结果]RT-PCR检测结果显示10Gy X射线照射48h后IEC-6细胞中PAI-1、IL-6mRNA表达明显升高[0.16∶0.76、0.20∶0.75(F=3.60,P=0.010)],10GyX射线+厄贝沙坦48h后明显降低[0.69∶0.46、0.77∶0.53(F=3.80,P=0.010)]。MTT结果显示10GyX射线照射会抑制IEC-6细胞的增殖,1、10、50μmol/L厄贝沙坦均促进IEC-6细胞照射后的增殖[0.38∶0.52、0.53、0.54(F=2.50,P=0.005)]。[结论]X射线照射IEC-6细胞后炎症因子IL-6、PAI-1mRNA表达升高。厄贝沙坦降低了IL-6、PAI-1mRNA的高表达,并促进了肠上皮细胞的增殖。
[Objective] To investigate the effects of Irbesartan, an angiotensin Ⅱ receptor antagonist, on the expression of plasminogen inhibitor-1 (PAI-1), interleukin- 6 (IL-6) mRNA expression and IEC-6 proliferative activity. [Method] The expression of angiotensin-converting enzyme 1 receptor (AT1 receptor) mRNA and AT2 receptor mRNA in IEC-6 cells was confirmed by RT-PCR. The expression of PAI-1 and IL-6 mRNA in IEC-6 cells treated with 10 Gy X-ray was observed by RT-PCR. The effect of Irbesartan and Irbesartan on the expression of PAI-1 and IL-6 mRNA in IEC-6 cells was also observed. MTT assay was used to observe the effect on cell proliferation. [Result] The results of RT-PCR showed that the expression of PAI-1 and IL-6 mRNA in IEC-6 cells significantly increased after irradiation with 10Gy X-ray 48h [0.16-0.76,0.20:0.75 (F = 3.60, P = 0.010) 10Gy X ray + irbesartan significantly reduced after 48h [0.69: 0.46,0.77: 0.53 (F = 3.80, P = 0.010)]. The results of MTT showed that 10Gy X-ray irradiation could inhibit the proliferation of IEC-6 cells. Irbesartan at 1, 10 and 50μmol / L promoted the proliferation of IEC-6 cells [0.38:0.52,0.53,0.54 (F = 2.50, P = 0.005)]. [Conclusion] The expression of IL-6 and PAI-1 mRNA in IEC-6 cells increased after X-ray irradiation. Irbesartan reduced the high expression of IL-6 and PAI-1 mRNA and promoted the proliferation of intestinal epithelial cells.
