目的评估图像细胞仪(ICM)所观察的几项参数用以揭示肝细胞癌(HCC)形成门静脉癌栓(TTPV)的价值。方法99例HCC以CAS┐200型ICM行DNA定量分析。结果DNA指数(DI)和增殖指数(PI)在合并TTPV的HCC分别显著高于无TTPV的HCC(P<0.05,P<0.01)。在同为二倍体的HCC中,合并TTPV者PI值显著高于无TTPV者(P<0.05)。合并TTPV的HCC,二倍体肿瘤的比例低于无TTPV者,而异倍体比例、细胞核面积、超5倍体细胞比例均高于无TTPV的HCC,但P>0.05。结论随着DNA合成水平的上升,HCC细胞获得了侵袭门静脉、形成TTPV的能力;以ICM行DNA定量分析对揭示HCC是否具有较强侵袭性有明显价值,其中PI是较为敏感的参数。
Objective To evaluate several parameters observed by image cytometry (ICM) to reveal the value of hepatocellular carcinoma (HCC) formation of portal vein tumor emboli (TTPV). METHODS: 99 cases of HCC were quantitatively analyzed by CAS-200 ICM. Results The DNA index (DI) and proliferation index (PI) of HCC with TTPV were significantly higher than those without TTPV (P<0.05, P<0.01). In the same diploid HCC, the PI value of patients with TTPV was significantly higher than those without TTPV (P<0.05). The proportion of diploid tumors in HTC with TTPV was lower than that in patients without TTPV, while the ratio of aneuploid, nuclei, and ultra-5-fold somatic cells were higher than those without TTPV, but P>0.05. Conclusion With the increase of DNA synthesis, HCC cells have the ability to invade the portal vein and form TTPV. Quantitative analysis of DNA by ICM has significant value in revealing whether HCC has strong invasiveness, and PI is a more sensitive parameter.
