聚丙烯酰胺凝胶等电聚焦技术,是目前分离蛋白质、两性高分子化合物的高分辨率方法之一。但两性电解质载体对染色有干扰,而且脱色时间过长。我们采用聚丙烯酰胺凝胶电聚焦法分离正常人血清,它可在两性电解质载体存在下直接染色,染色时间7分钟,脱色3小时左右,背景着色浅,蛋白带清楚,灵敏度高。本方法既大大缩短了凝胶电聚焦过程的时间;又避免了某些物质的干扰,有利于我们进一步探索聚丙烯酰胺凝胶这一高分辨方法用于分离提纯肝癌抗原及其染色,将为缩短整个实验过程,提高分辨率打下了基础。
Polyacrylamide gel isoelectric focusing technology is one of the high resolution methods for separating proteins and amphoteric polymer compounds. However, the ampholyte carrier interferes with dyeing and the decolorization time is too long. We used a polyacrylamide gel electrofocusing method to separate normal human serum. It can be directly stained in the presence of amphoteric carriers, with a staining time of 7 minutes, decoloration of about 3 hours, light background color, clear protein bands, and high sensitivity. This method not only greatly shortens the time of the gel electrofocusing process, but also avoids the interference of certain substances. This helps us to further explore the high-resolution method of polyacrylamide gels for the separation and purification of liver cancer antigens and their staining. Shorten the entire experimental process and lay a foundation for improving resolution.
