目的 研究小剂量牛初乳短链胰岛素样生长因子 Ⅰ (Bc tIGF Ⅰ )、重组人IGF Ⅰ(rhIGF Ⅰ )对链脲佐菌素 (STZ)诱导的糖尿病大鼠周围神经病变的作用。方法 早期给STZ糖尿病大鼠腹腔注射Bc tIGF Ⅰ和rhIGF Ⅰ (均为 75 0ng·kg-1·d-1)。 11周后对各组大鼠进行神经电生理及病理形态分析。结果 Bc tIGF Ⅰ治疗后坐骨神经运动神经传导速度、动作电位及H反射潜伏期都明显改善 (P <0 .0 5 ) ,神经病理也明显改善。rhIGF Ⅰ的治疗对坐骨神经运动传导速度、波幅及H反射潜伏期没有影响 (P >0 .0 5 ) ,神经病理也没有明显改善。结论 小剂量Bc tIGF Ⅰ (75 0ng·kg-1·d-1)能改善糖尿病神经病变 ,而同等剂量的rhIGF Ⅰ无此作用。
Objective To investigate the effect of short-term insulin-like growth factor Ⅰ (Bc tIGF Ⅰ) and recombinant human IGF-Ⅰ (Ⅰ) on the peripheral neuropathy induced by streptozotocin (STZ) in diabetic rats. Methods Bc tIGF Ⅰ and rhIGF Ⅰ (75 0 ng · kg -1 · d -1) were injected intraperitoneally into STZ diabetic rats. Eleven weeks later, the electrophysiological and pathological analysis of the rats in each group were performed. Results The nerve conduction velocity, action potential and H reflex latency of sciatic nerve were significantly improved after Bc tIGF Ⅰ treatment (P <0.05), and the neuropathology was significantly improved. The treatment of rhIGF Ⅰ had no effect on motor conduction velocity, amplitude and latency of H reflex (P> 0.05), and neuropathology did not improve significantly. Conclusion Low dose Bc tIGF Ⅰ (75 0 ng · kg -1 · d -1) can improve the diabetic neuropathy, while the same dose of rhIGF Ⅰ does not.
