目的优化极地真菌S-3-66-E1转化北沙参中总香豆素的条件。方法采用静息细胞转化法,考察了生物转化条件(菌体密度、底物浓度、温度、pH、摇床转速、金属离子)对转化体系的影响,并通过正交实验优化工艺参数,利用HPLC检测总香豆素的转化情况。结果确定摇瓶转化的最优条件:培养温度15℃,摇床转速240r/min,菌体密度60g·L~(-1),底物浓度0.1g·L~(-1),pH值5.9,转化时间24h,在此条件下总香豆素的转化量趋近完全。结论采用静息细胞法转化北沙参中总香豆素的工艺稳定可行。
Objective To optimize the conditions for conversion of total coumarin from Psathyrostachys spp. Methods The effects of biotransformation conditions (cell density, substrate concentration, temperature, pH, shaking speed and metal ions) on the transformation system were investigated by resting cell transformation method. The technological parameters were optimized by orthogonal experiments. Test coumarin conversion. The results showed that the optimum conditions for shake flask transformation were: culture temperature 15 ℃, shaking speed 240r / min, cell density 60g · L -1, substrate concentration 0.1g · L -1, pH 5.9 , The conversion time 24h, under the conditions of total coumarin conversion is almost complete. Conclusion The process of transforming total coumarins in Bei Sha ginseng by resting cell method is stable and feasible.
