目的:建立高效液相色谱法测定无梗五加果实微波消解后齐墩果酸含量的方法。方法:采用微波消解法对无梗五加果实中的齐墩果酸苷类成分进行水解,并采用高效液相色谱法测定苷元齐墩果酸的含量,色谱条件为Promosil C18色谱柱(4.6 mm×250 mm,5μm),流动相甲醇-0.2%乙酸水溶液(90∶10),流速1.0 m L·min-1,检测波长210 nm。结果:消解的条件为9%的盐酸,0.1 MPa微波消解0.5 h;齐墩果酸的线性范围是0.781 6~3.908μg(r=0.999 6),平均回收率为97.3%(RSD1.2%)。结论:该方法准确、可靠,可用于无梗五加果实的质量控制。
Objective: To establish a method for the determination of oleanolic acid in microwave-digested non-stem five-fruits by high performance liquid chromatography. Methods: Oleanolic acid glycoside was hydrolyzed by microwave digestion method. The content of oleanolic acid was determined by HPLC. Promosil C18 column (4.6 mm × 250 mm, 5 μm), mobile phase methanol-0.2% acetic acid aqueous solution (90:10), flow rate 1.0 mL · min-1 and detection wavelength 210 nm. Results: The conditions of digestion were 9% hydrochloric acid and 0.1 MPa microwave digestion for 0.5 h. The linear range of oleanolic acid was 0.781 6 ~ 3.908 μg (r = 0.999 6) with an average recovery of 97.3% (RSD 1.2%). . Conclusion: This method is accurate and reliable, and can be used for quality control of the fruit of Acanthopanax senticosus.
