目的:探讨氯化钾(KCl)所致大鼠肾上腺嗜铬细胞瘤细胞(PC12)内钙超载对其细胞活性及磷酸化cAMP反应原件绑定蛋白(p-CREB)表达的影响。方法:将PC12细胞随机分为对照组与干预组,对照组不作任何药物干预,干预组予以KCl 100 mmol/L干预10 min。用MTT法检测两组细胞活性,应用Western blot检测PC12细胞内蛋白激酶A(PKA)活性及p-CREB的表达。结果:MTT法检测细胞活性,KCl组较对照组细胞活性下降,有统计学意义(P<0.05);应用Western blots检测,KCl组PKA活性较对照组明显降低;KCl组p-CREB表达较对照组明显降低。结论:KCl所致钙超载对PC12活性有明显的毒性作用,降低PKA的活性,减少CREB的激活。
OBJECTIVE: To investigate the influence of calcium overload in rat adrenal pheochromocytoma (PC12) cells induced by potassium chloride (KCl) on its cell viability and expression of phosphorylated cAMP response element binding protein (p-CREB). Methods: PC12 cells were randomly divided into control group and intervention group. The control group received no drug intervention. The intervention group was treated with KCl 100 mmol/L for 10 min. The cell viability of the two groups was detected by MTT assay. The activity of protein kinase A (PKA) and the expression of p-CREB in PC12 cells were detected by Western blot. Results: The cell viability was detected by MTT assay. The activity of KCl group was lower than that of the control group (P<0.05). The PKA activity in KCl group was significantly lower than that in the control group by Western blots; the expression of p-CREB in KCl group was lower than that in the control group. The group is significantly lower. CONCLUSION: Calcium overload caused by KCl has obvious toxic effects on PC12 activity, which decreases the activity of PKA and reduces the activation of CREB.
