本研究用纯化E.coliRecA蛋白免疫BALB/c小鼠,取免疫脾细胞与小鼠骨髓瘤细胞Sp2/0融合,以粗制E.coliRecA包被酶标反应板,间接ELISA方法筛选阳性孔,经有限稀释法4次克隆化,建立了两株分泌抗RecA蛋白McAb的细胞系2D6和3B1。免疫印迹表明McAb只对细菌的RecA有反应,不与细菌其它成分发生反应。间接ELISA进一步表明,McAb2D6、3D1只对细菌RecA类蛋白有反应,不与小鼠肝、心、肺、脾等和BHK细胞的内外蛋白反应。
This study used purified E. The BALB / c mice were immunized with the coliRecA protein, and the splenocytes were fused with Sp2 / 0 of mouse myeloma cells. The positive clones were screened by indirect enzyme-linked immunosorbent assay (ELISA) and cloned by restriction endonuclease four times. Two cell lines, 2D6 and 3B1, secreting anti-RecA McAb were established. Western blotting showed that McAb reacted only to bacterial RecA and did not react with other bacterial components. Indirect ELISA further showed that McAb2D6,3D1 only reacted with bacterial RecA protein and did not react with the internal and external proteins of mouse liver, heart, lung, spleen and other BHK cells.
