目的:研究何首乌总苷(polygonummultiflorumtotalglycoside熏PMTG)对载脂蛋白E基因缺陷(apoE-蛐-)小鼠实验性动脉粥样硬化(atherosclerosis,As)病变血脂及抗氧化能力的影响。方法:将apoE-蛐-小鼠随机分为4组:何首乌总苷高剂量组(150mg蛐kg蛐d)、低剂量组(25mg蛐kg蛐d)、阿托伐他汀阳性药物组(5mg蛐kg蛐d)及空白对照组,给药第10周后全部处死,酶法检测四组血清脂质含量,化学法测氧化指标一氧化氮(NO)、总抗氧化能力(TAC)及丙二醛(MDA)。结果:实验10周后,何首乌总苷高、低剂量组及阳性药物组与空白对照组比较,显示(1)何首乌总苷可以显著降低血清总胆固醇(TC)、甘油三酯(TG)水平,升高高密度脂蛋白-胆固醇(HDL-C)水平(P<0.01);(2)明显增加血浆NO及TAC(P<0.05及P<0.01)熏减少MDA的生成(P<0.01)。结论:何首乌总苷可以调节apoE-蛐-小鼠血脂代谢、增强其抗氧化能力,起到防止apoE-蛐-小鼠实验性动脉粥样硬化病变形成的作用。
OBJECTIVE: To study the effect of Polygonum multiflorum total glycolside smoked PMTG on blood lipids and antioxidant capacity of experimental atherosclerosis (As) lesions in apolipoprotein E-deficient (apoE-蛐-) mice. METHODS: ApoE-蛐-mice were randomly divided into 4 groups: high dose group of total glycosides of Polygonum multiflorum (150 mg/kg/day), low dose group (25 mg/kg/day), and atorvastatin positive drug group (5 mg/day). Kg蛐d) and blank control group. All rats were sacrificed after the 10th week of administration. The serum lipid contents of the four groups were measured by enzyme method, and the oxidation index (NO), total antioxidant capacity (TAC) and propane II were measured by chemical method. Aldehyde (MDA). Results: Compared with the control group, the total glucosides of high dose, low dose group and positive drug group of Polygonum multiflorum in 10 weeks after the experiment showed that (1) total glycosides of Polygonum multiflorum can significantly reduce serum total cholesterol (TC) and triglyceride (TG) levels. Increased high-density lipoprotein-cholesterol (HDL-C) levels (P <0.01); (2) significantly increased plasma NO and TAC (P <0.05 and P <0.01) smoked reduced MDA production (P <0.01). Conclusion: The total glycosides of Polygonum multiflorum Thunb. can regulate the lipid metabolism of apoE-蛐-mice and enhance its antioxidative capacity. It can prevent the formation of experimental atherosclerotic lesions in apoE-蛐-mice.
