目的研究甲状腺功能低下对新生期鼠脑梨状皮质Goα及GsαmRNA表达的影响。 方法采用地高辛标记寡核苷酸探针原位杂交技术 ,对围生期正常及甲减Wistar大鼠动物模型梨状皮质Goα及GsαmRNA的表达状况进行研究。 结果 14d龄甲减鼠脑梨状皮质GoαmRNA的表达明显升高 ,但其GsαmRNA与对照组无显著差异。结论在鼠脑发育过程中 ,甲状腺激素对梨状皮质Goα基因的表达具有负调作用 ,而对该区Gsα基因的表达则不具备调节功能
Objective To investigate the effect of hypothyroidism on piriform cortex Goα and GsαmRNA expression in neonatal rats. Methods Digoxin-labeled oligonucleotide probe in situ hybridization was used to study the expression of piriform cortex Goα and GsαmRNA in normal and hypothyroidism Wistar rats during perinatal period. Results The expression of Goα mRNA in cortical pyramidal cortex of 14-day-old hypothyroidism rats was significantly increased, but there was no significant difference between GsαmRNA and control group. Conclusion Thyroid hormone plays a negative role in the expression of Goα gene in piriform cortex during the development of rat brain and does not regulate the expression of Gsα gene
