目的观察白介素-21(IL-21)和白介素-2(IL-2)联合诱导的人外周血淋巴细胞(PBMC)抗卵巢癌细胞OVCAR-3的作用。方法收集河南省人民医院2015年1月至2016年1月健康志愿者外周血,密度梯度离心法分离人PBMC,分别用IL-2、IL-21或IL-2联合IL-21联合诱导培养,观察外周血中淋巴细胞增殖、细胞表型,并检测诱导的淋巴细胞对卵巢癌细胞株的杀伤作用。结果 IL-21处理组的淋巴细胞浓度高于IL-2单独处理组,IL-21具有较强的CD3CD56细胞亚群诱导作用,但两者联合并不能增进其促增殖和诱导CD3CD56亚群的作用。IL-21和IL-2联合诱导的淋巴细胞对OVCAR-3抑制率和促凋亡率最好,优于单用IL-2或IL-21。结论 IL-21和IL-2联合能增加外周血淋巴细胞杀伤卵巢癌细胞OVCAR-3的作用。
Objective To observe the effect of human peripheral blood lymphocyte (PBMC) induced by interleukin-21 (IL-21) and interleukin-2 (IL-2) on ovarian cancer cell line OVCAR-3. Methods Peripheral blood from healthy volunteers from January 2015 to January 2016 in Henan Provincial People’s Hospital was collected. Human peripheral blood mononuclear cells (PBMCs) were isolated by density gradient centrifugation and cultured with IL-2, IL-21 or IL-2, Peripheral blood lymphocyte proliferation and cell phenotype were observed, and the killing effect of induced lymphocytes on ovarian cancer cell lines was detected. Results The IL-21-treated group had higher lymphocyte concentration than IL-2 alone group, and IL-21 had a stronger induction of CD3CD56 cell subsets. However, the combination of both could not promote the proliferation and induce the CD3 CD56 subsets . Lymphocytes induced by IL-21 and IL-2 had the best effect on OVCAR-3 inhibition and apoptosis, which was better than that of IL-2 or IL-21 alone. Conclusion The combination of IL-21 and IL-2 can increase the effect of peripheral blood lymphocyte killing ovarian cancer cell line OVCAR-3.
