目的研究大环内酯类药物对哮喘大鼠肺组织窖蛋白-1(Cav-1)和凋亡相关蛋白B细胞淋巴瘤/白血病-2(Bcl-2)蛋白表达的影响及二者的相互关系。方法将40只大鼠随机分为4组:对照组、模型组、红霉素干预组(A1组)、红霉素+地塞米松干预组(A2组)。以卵白蛋白致敏制作大鼠哮喘模型。应用免疫组织化学法和蛋白印迹方法检测不同组别大鼠肺组织Cav-1和Bcl-2蛋白的表达。结果免疫组织化学法和蛋白印迹方法检测不同组别大鼠肺组织Cav-1和Bcl-2蛋白表达结果一致。模型组大鼠肺组织Cav-1蛋白表达显著低于其他各组(Pa<0.01)。A2组大鼠肺组织Cav-1蛋白表达显著高于A1组(P<0.05),但A1组和A2组均显著低于对照组(Pa<0.01)。模型组大鼠肺组织Bcl-2蛋白表达显著高于对照组和A2组(P<0.05,0.01),但与A1组相比差异无统计学意义(P>0.05)。A1组、A2组与对照组相比,Bcl-2蛋白表达均显著升高(Pa<0.01),但A1组与A2组比较差异无统计学意义(P>0.05)。Cav-1与Bcl-2蛋白表达呈显著负相关(r=-0.618,P<0.01)。结论大环内酯类药物可协同糖皮质激素通过上调Cav-1表达和下调Bcl-2表达促进细胞凋亡,从而发挥抗炎作用。
Objective To investigate the effects of macrolide on the protein expression of Cav-1 and Bcl-2 in lung tissue of asthmatic rats and the relationship between the two relationship. Methods Forty rats were randomly divided into 4 groups: control group, model group, erythromycin intervention group (A1 group), erythromycin + dexamethasone intervention group (A2 group). Ovalbumin sensitized rat asthma model. Immunohistochemistry and Western blotting were used to detect the expression of Cav-1 and Bcl-2 in different groups of rats. Results Immunohistochemistry and Western blotting were used to detect the expression of Cav-1 and Bcl-2 protein in different groups of rats. Cav-1 protein expression in lungs of model rats was significantly lower than that of other groups (Pa <0.01). The expression of Cav-1 protein in lung tissue of A2 group was significantly higher than that of A1 group (P <0.05), but the levels of Cav-1 in group A1 and A2 were significantly lower than those in control group (Pa0.01). The expression of Bcl-2 protein in lung tissue of model group was significantly higher than that of control group and A2 group (P <0.05, 0.01), but there was no significant difference compared with A1 group (P> 0.05). Compared with control group, the expression of Bcl-2 protein in group A1 and group A2 were significantly increased (Pa <0.01), but there was no significant difference between group A1 and group A2 (P> 0.05). Cav-1 and Bcl-2 protein expression was significantly negatively correlated (r = -0.618, P <0.01). Conclusions Macrolides can play an anti-inflammatory role in collaboration with corticosteroids by up-regulating Cav-1 expression and down-regulating Bcl-2 expression.
