美国联合农业种子公司最近通过小孢子离体培养从单个单倍体细胞获得了玉米自交系。他们报道,当来自高度雄核发育基因型玉米的花药被诱导开裂,将释放的小孢子培养在液体诱导培养基中,每100个培养的花药能产生多达400个“胚状结构”,大约有5~25%的“胚状结构”能产生将来会再生植株的愈伤组织。利用这一系统,能获得大量来自小孢子的双单倍体。
United National Agricultural Seeds recently acquired maize inbred lines from single haploid cells by microspore in vitro culture. They reported that when anther from highly male and maize genotype maize was induced to dehiscence and the released microspores cultured in liquid induction medium, up to 400 “embryoid-like structures” could be produced per 100 cultured anthers, approximately Five to twenty-five percent of “embryoid-like structures” produce callus that will regenerate plants in the future. With this system, a large amount of doubled haploid from microspores can be obtained.
