采用增殖细胞核抗原(PCNA)免疫组化标记与图像分析技术,对30例人体肝细胞癌与癌旁近癌肝细胞的增殖活性进行了定量研究。结果显示:(1)癌与癌旁近癌肝细胞PCNA阳性平均标记率分别为45.22%±17.90%和12.74%±4.89%(P<0.01),随肝癌分级升高,PCNA标记享有升高的趋势。(2)40%以上肝癌细胞的PCNA相对含量与癌旁肝细胞PCNA平均含量相似,结合病理形态分析,认为癌细胞PCNA含量增高与低分化癌细胞核增大关系密切。(3)用图像分析技术显示的PCNA标记数密度与常规计数法所得PCNA标记率有较好一致性,且前者更为客观。
Using proliferating cell nuclear antigen (PCNA) immunohistochemistry and image analysis techniques, the proliferative activity of 30 cases of human hepatocellular carcinoma and adjacent cancerous hepatocytes was quantitatively studied. The results showed that: (1) The positive rate of PCNA positive in cancerous and adjacent cancerous hepatocytes were 45.22%±17.90% and 12.74%±4.89%, respectively (P<0.01). As grades increase, the PCNA marker has an increased tendency. (2) The relative content of PCNA in hepatoma cells with 40% or more was similar to the average PCNA content in adjacent liver cells. Combined with histopathological analysis, it was considered that the increase in PCNA content in cancer cells was closely related to the increase in the number of poorly differentiated cancer cells. (3) The number density of PCNA markers displayed by image analysis technique is in good agreement with the PCNA labeling rate obtained by conventional counting methods, and the former is more objective.
