萝卜胞质雄性不育(OguCMS)是目前甘蓝中应用较广的雄性不育类型,MYB转录因子具有调控植物防御应答反应和多个发育过程的作用。本实验以在甘蓝(Brassica oleracea var.capitata)OguCMS花药中下调10.3倍的EST序列为信息探针,结合电子克隆及RACE技术,得到一个与甘蓝OguCMS雄性不育相关的MYB转录因子全长cDNA,命名为BoMYB1(GenBank登录号:JN703995)。经亚细胞定位预测,该基因定位于细胞核,全长1141bp,包含一个长度为196bp的5’非翻译区、246bp的3’非翻译区和一个699bp的开放阅读框。该基因在花药中具表达优势,并在花药发育晚期出现表达高峰,受植物激素水杨酸(SA)和茉莉酸甲酯(JA-ME)的调控,诱导花药发育基因的表达。实验结果提示,BoMYB1可能是参与OguCMS花药发育的重要基因之一。
Radish cytoplasmic male sterility (OguCMS) is a genus of male sterility widely used in cabbage. MYB transcription factor plays a role in regulating plant defense response and multiple developmental processes. In this study, a full-length cDNA of MYB transcription factor related to male sterility of OguCMS in Brassica oleracea var.capitata Oryza sativa was obtained by using a 10.3-fold-down-regulated EST sequence as probe. Named BoMYB1 (GenBank Accession Number: JN703995). The subcellular localization predicts that the gene is located in the nucleus and is 1141bp in length. It contains a 5 ’untranslated region of 196bp, a 3’ untranslated region of 246bp and a 699bp open reading frame. The gene has the advantage of expression in anther and its expression peak appears in late stage of anther development. It is regulated by plant hormone salicylic acid (SA) and methyl jasmonate (JA-ME) to induce the expression of anther development genes. The experimental results suggest that BoMYB1 may be one of the important genes involved in the development of anther in OguCMS.
