目的 探讨树突状细胞 (DC)在 5 / 6肾切除大鼠肾小管间质中分布及其与肾间质纤维化的关系。方法 5 4只SD大鼠均分为正常组、假手术组、模型组 (切除 5 / 6肾 ,建立慢性肾功能衰竭模型 )。分别于建模第1、4和 12周 ,采用免疫双标记染色及荧光图像分析法 ,观察CD1a +CD80 +DC在各组大鼠肾组织中的分布变化 ;采用免疫组织化学及半定量分析 ,观察黏附分子P 选择素以及转化生长因子 (TGF) β1、结缔组织生长因子 (CT GF)、α 平滑肌肌动蛋白 (α SMA)、Ⅲ型胶原 (ColⅢ )、纤维连接蛋白 (FN)在上述肾组织中的表达改变 ;此外采用Masson染色评定各组肾小管间质纤维化 (TIF)程度。结果 ①模型组CD1a +CD80 +DC主要分布于肾小管、肾间质和肾血管 ,以肾间质最为明显 ;其分布数量于建模后第 12周较第 1、4周明显增多 (P <0 .0 1) ,且与肾功能指标血尿素氮 (BUN)、血肌酐 (SCr)、内生肌酐清除率 (Ccr)明显相关 (r值分别为 0 .2 5 4、0 .2 6 3、- 0 .36 7,P值<0 .0 5~ 0 .0 0 1) ;② 12周时 ,P 选择素、TGF β1、CTGF、α SMA、ColⅢ、FN以肾小管间质为主表达以及TIF程度均明显增加 ,并与CD1a +CD80 +DC分布数量成正相关 (r值分别为 0 .5 6 6、0 .4 77、0 .4 2 5、0 .2 88、0 .4 0 8、0 .4 95、0 .82 3,P值 <0
Objective To investigate the distribution of dendritic cells (DCs) in the tubulointerstitial of 5/6 nephrectomized rats and its relationship with renal interstitial fibrosis. Methods 5 4 SD rats were divided into normal group, sham operation group and model group (5/6 nephrectomy, chronic renal failure model). Immunocytochemical staining and semi-quantitative analysis were used to observe the distribution of CD1a + CD80 + DC in renal tissues of rats in each group at the 1st, 4th, and 12th week of modeling. The expression of P-selectin, transforming growth factor (TGF) β1, connective tissue growth factor (CTGF), α-SMA, Col Ⅲ and fibronectin The expression of TIF in the tissue was also observed. In addition, Masson staining was used to evaluate the degree of tubulointerstitial fibrosis (TIF). Results ① The distribution of CD1a + CD80 + DC in the model group was mainly in the renal tubule, renal interstitium and renal artery, and the most obvious was in the renal interstitium.
