为了研究根皮素对LPS诱导的小鼠乳腺组织的炎症变化,并证明根皮素对乳腺炎具有保护作用,试验应用乳导管内灌注LPS建立小鼠乳腺炎模型,进行H.E.染色、ELISA检测、Western-blott检测和髓过氧化物酶(MPO)检测等研究根皮素对乳腺组织的保护作用。结果表明:LPS能使乳腺组织产生病理学损伤,促进白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的产生,增加组织MPO活性,增强NF-κB信号通路蛋白的表达,而根皮素能够抑制LPS诱导的小鼠乳腺组织的炎症变化。说明根皮素对LPS诱导的小鼠乳腺炎具有一定保护作用。
In order to study the changes of inflammatory response induced by phloretin in mouse mammary gland induced by LPS and to prove that phloretin has a protective effect on mastitis, the model of mouse mastitis was established by intraperitoneal injection of LPS in dairy cow. HE staining, ELISA, Western-blott test and myeloperoxidase (MPO) test of phloretin on breast tissue protection. The results showed that LPS could induce the pathological damage of breast tissue and promote the production of interleukin-1β, interleukin-6 and tumor necrosis factor-α (TNF-α) MPO activity was increased and the expression of NF-κB signaling pathway protein was enhanced. Phloretin could inhibit LPS-induced inflammation in mouse mammary gland tissue. Phloretin on LPS-induced mouse mastitis has a protective effect.
