目的探讨黏蛋白1(MUC1)在大鼠呼吸道内的分布以及在哮喘大鼠模型的表达水平的改变。方法将40只Wistar大鼠随机分为正常对照组(n=20)和哮喘模型组(n=20),采用卵清蛋白(OVA)致敏法建立支气管哮喘的大鼠模型。应用免疫组织化学法和蛋白印迹法测定MUC1蛋白在正常大鼠呼吸道内的分布以及在哮喘大鼠中的变化特点。结果 MUC1蛋白在正常对照组呼吸道内分布在气道上皮纤毛细胞内,而在毗邻的无纤毛细胞内MUC1蛋白为阴性;在哮喘模型组纤毛细胞内MUC1蛋白阳性表达增强,MUC1蛋白量较正常对照组增多约36%(P<0.05)。结论 MUC1在气道上皮内分布,可能对气道起保护、润滑作用;在哮喘发病过程中,MUC1蛋白表达量增加可能与MUC1免疫调节功能相关。
Objective To investigate the distribution of mucin 1 (MUC1) in the respiratory tract of rats and the changes in the expression of MUC1 in asthmatic rats. Methods Forty Wistar rats were randomly divided into normal control group (n = 20) and asthma model group (n = 20). The rat model of bronchial asthma was established by ovalbumin (OVA) sensitization. Immunohistochemistry and Western blotting were used to determine the distribution of MUC1 protein in the respiratory tract of normal rats and its changes in asthmatic rats. Results MUC1 protein was distributed in the airway epithelial ciliated cells in the respiratory tract of normal control group, while MUC1 protein was negative in adjacent ciliated cells. MUC1 protein expression was increased in ciliated cells of asthma model group, The group increased by about 36% (P <0.05). Conclusion The distribution of MUC1 in the airway epithelium may play a protective and lubricating role on the airway. MUC1 protein expression may be related to MUC1 immunoregulatory function in the pathogenesis of asthma.
