目的 分析柯萨奇 B3病毒 (CVB3 )中国分离株编码主要中和抗原的 VP1基因序列。方法 用逆转录 PCR技术扩增 CVB3中国分离株的 VP1基因 ,通过 TA克隆法构建 p CR2 .1-VP1并进行核苷酸序列的测定分析。结果 测序发现 CVB3中国分离株与 CVB3 Nancy株 VP1基因均编码 2 93个氨基酸 ,二者存在 5 9个核苷酸差异 ,其中 10处影响到氨基酸的编码 ,其余均为同义变异。结论 CVB3中国分离株与 Nancy株 VP1基因序列存在一定的差异 ,该差异对免疫学性状的影响有待进一步鉴定
Objective To analyze the VP1 gene sequence encoding the major neutralizing antigen of Chinese isolate of Coxsackie B3 virus (CVB3). Methods The VP1 gene of CVB3 Chinese isolate was amplified by reverse transcription polymerase chain reaction (RT-PCR). The p CR2 .1-VP1 was constructed by TA cloning and the nucleotide sequence was analyzed. Results The sequencing results showed that the VP1 gene of CVB3 strain and CVB3 Nancy strain all encoded 293 amino acids. There were 59 nucleotide differences between the two strains, of which 10 affected the amino acid coding and the others were synonymous. Conclusion The sequence of VP1 gene between CVB3 isolate and Nancy strain is different, and the effect of this difference on immunological traits needs to be further identified
