目的 探讨虫草多糖对大鼠肝星状细胞的作用机制。方法 分离大鼠肝星状细胞 ,采用不同浓度的虫草多糖处理后 ,分别应用MTT法、凝胶迁移率法、ELISA法和Northernblot检测大鼠肝星状细胞的增殖、核因子κB(NF κB)活性和肿瘤坏死因子 α蛋白和mRNA的表达。结果 虫草多糖对大鼠肝星状细胞生长具有较强的抑制作用。虫草多糖可明显抑制大鼠肝星状细胞核因子κB活性和下调肿瘤坏死因子 α蛋白和mRNA的表达 ,并且呈剂量依赖性。结论 虫草多糖对大鼠HSC增殖具有很强的抑制作用 ,可能与抑制NF κB活性和下调肿瘤坏死因子 α表达有关
Objective To investigate the mechanism of Cordyceps polysaccharide on rat hepatic stellate cells. METHODS Rat hepatic stellate cells were isolated and treated with different concentrations of Cordyceps polysaccharides. The proliferation of rat hepatic stellate cells and nuclear factor kappa B (NF κB) were detected by MTT assay, gel mobility assay, ELISA and Northern blot. Activity and expression of tumor necrosis factor alpha protein and mRNA. Results Cordyceps polysaccharide had a strong inhibitory effect on the growth of rat hepatic stellate cells. Cordyceps polysaccharide can significantly inhibit the activity of rat NF-κB and down-regulate the expression of TNF-α protein and mRNA in a dose-dependent manner. Conclusion Cordyceps polysaccharide has a strong inhibitory effect on rat HSC proliferation, which may be related to the inhibition of NF-κB activity and down-regulation of tumor necrosis factor α expression.
