目的 观察从出生至50天的大鼠视皮层中N-甲基-D-天门冬氨酸受体(N-methyl-D-aspartate receptor, NMDAR)功能亚单位NR2B的表达变化,为研究视觉刺激对视皮层NR2B的表达调控机制作准备。设计 实验性研究。研究对 象 30只新生正常健康纯种Wistar大鼠。方法 30只大鼠随机分为10组,每组3只,正常饲养,分别于出生后第0、5、10、15、 20、25、30、35、40、50天处死,大鼠脑视皮层行30μm厚冠状冰冻切片,应用免疫组织化学链霉亲和素-生物素-过氧化物酶复合 物(streptomycin-avidin-biotin-peroxidase complex,SABC)法显色,显微镜观察NR2B的表达情况并进行图像分析。主要指标 平均光密度(average optical density,AOD)与综合光密度(integrated optical density,IOD)。结果 NR2B在出生时的视皮层中 即已存在,之后呈逐渐上升趋势,至出生后第15天达到峰值,继而逐渐下降,至第50天趋于平稳。结论 在大鼠出生后0-15 天,NR2B水平呈逐渐上升状态,提示NR2B对出生后大鼠的视觉发育有着重要的生理作用。自第15天大鼠睁眼起,NR2B水平 逐渐下降,表明NR2B的弱化表达与视觉刺激密切相关。
Objective To observe the expression changes of N-methyl-D-aspartate receptor (NMDAR) subunit NR2B in rat visual cortex from birth to 50 days. Prepare the mechanism of NR2B expression in the visual cortex. Design experimental research. Thirty new normal healthy Wistar rats were included in the study. Methods Thirty rats were randomly divided into 10 groups with 3 rats in each group. The rats were reared normally and sacrificed at 0, 5, 10, 15, 20, 25, 30, 35, 40 and 50 days after birth. The cornea was frozen in 30 μm thick cortex. The expression of NR2B was observed by immunohistochemistry streptavidin-biotin-peroxidase complex (SABC) Perform image analysis. The main indicators of the average optical density (average optical density, AOD) and integrated optical density (integrated optical density, IOD). Results NR2B was present in the visual cortex at birth, and then gradually increased. It peaked on the 15th day after birth and then decreased gradually until the 50th day. Conclusion At 0-15 days after birth, the level of NR2B in rats gradually increases, suggesting that NR2B has an important physiological role in the visual development of rats after birth. Since the 15th day when rats opened their eyes, the level of NR2B gradually decreased, indicating that the weakened expression of NR2B is closely related to visual stimulation.
