目的观察银杏酮酯(EGb_(50))对大鼠坐骨神经损伤后神经中诱导型一氧化氮合酶(iNOS)表达的影响。方法 SD 大鼠156只,随机分成假手术组、损伤对照组与实验组,后两组切断右侧坐骨神经并缝合。实验组给予 EGb_(50) 200 mg·kg~(-1)·d~(-1)溶于1 ml 生理盐水中灌胃,损伤对照组给予生理盐水1 ml 灌胃,假手术组仅显露右侧坐骨神经,术后不作处理。分别于术后1、3、7、14、21及28 d 取吻合口远端的神经,采用免疫组织化学和实时荧光定量聚合酶链反应(PCR)方法检测损伤神经组织中 iNOS 的表达水平。结果实验组 iNOS 阳性染色吸光度均值(A)在术后3、7、14和21 d均明显低于对照组(均 P<0.05),术后1、3和7 d,实验组坐骨神经中 iNOS mRNA 的表达均低于对照组(P<0.01)。结论银杏酮酯可抑制大鼠坐骨神经损伤后神经纤维中 iNOS 的表达,其促进神经再生的作用机制可能与抑制 iNOS 的表达有关。
Objective To observe the effect of EGb_(50) on the expression of inducible nitric oxide synthase (iNOS) in the nerves of rats after sciatic nerve injury. Methods 156 SD rats were randomly divided into sham operation group, injury control group and experimental group. The right sciatic nerve was cut and sutured in the latter two groups. The experimental group was given intragastric administration of EGb 50 with 200 mg·kg -1 ·d -1 dissolved in 1 ml normal saline. The control group was given intragastric administration of 1 ml of saline, and the sham group showed only right. Lateral sciatic nerve, no postoperative treatment. The nerves at the distal end of the anastomosis were taken at 1,3,7,14,21, and 28 days after operation. The expression of iNOS in injured nerve tissue was detected by immunohistochemistry and real-time fluorescence quantitative polymerase chain reaction (PCR). Results The average absorbance of iNOS positive staining in the experimental group was significantly lower than that in the control group on the 3rd, 7th, 14th, and 21st postoperative days (all P<0.05). On the 1st, 3rd, and 7th days after the operation, iNOS mRNA in the sciatic nerve of the experimental group was detected. The expression was lower in the control group (P<0.01). Conclusion Ginkgo Biloba extract can inhibit the expression of iNOS in nerve fibers after sciatic nerve injury in rats, and its mechanism of promoting nerve regeneration may be related to the inhibition of iNOS expression.
