目的:探讨乳腺浸润性导管癌组织中p27、Skp2和E-cad的表达及其生物学意义。方法:采用微阵分析免疫组化法检测p27、Skp2和E-cad在浸润性导管癌组织、淋巴结转移灶与乳腺良性病变中的表达强度。结果:浸润性导管癌组织中p27和E-cad的表达强度低于乳腺良性病变,χ2分别为13.648、63.163,P值均<0.05;Skp2的表达强度高于乳腺良性病变,χ2=17.150,P<0.05。在浸润性导管癌组织中Skp2+/p27低指数与核异形性、组织学分级和癌巢分化程度及淋巴结转移情况具有相关性,χ2分别为12.634、6.397、8.750、23.768,P值均<0.05,与间质纤维化程度、年龄和肿物大小无相关性,χ2分别为1.325、0.281、3.748,P值均>0.05。浸润性导管癌淋巴结转移灶的E-cad表达强度低于原发瘤,χ2=23.142,P<0.05。结论:p27、Skp2和E-cad在乳腺肿瘤中与肿瘤分化程度、组织学分级和肿瘤转移有关。Skp2+/p27低指数和E-cad表达强度可能对乳腺癌患者的诊断及预后评估有意义。
Objective: To investigate the expression of p27, Skp2 and E-cad in breast invasive ductal carcinoma and its biological significance. Methods: The expression of p27, Skp2 and E-cad in invasive ductal carcinoma tissues, lymph node metastasis and benign breast lesions were detected by using microarray immunohistochemistry. Results: The expression of p27 and E-cad in invasive ductal carcinoma was lower than that in benign breast lesions (χ2 = 13.648,63.163, P <0.05). The expression of Skp2 was higher than that in benign breast, χ2 = 17.150, P <0.05. In infiltrating ductal carcinoma, the low index of Skp2 + / p27 correlated with nuclear atypia, histological grade, cancer nest differentiation and lymph node metastasis (χ2 = 12.634,6.397,8.750,23.768, P <0.05 respectively) There was no correlation with the degree of interstitial fibrosis, age and size of tumor, χ2 = 1.325,0.281,3.748, P> 0.05 respectively. The invasiveness of invasive ductal carcinoma with lymph node metastasis E-cad expression intensity lower than the primary tumor, χ2 = 23.142, P <0.05. Conclusion: The expression of p27, Skp2 and E-cad in breast cancer is related to tumor differentiation, histological grade and tumor metastasis. Skp2 + / p27 low index and E-cad expression may be useful in the diagnosis and prognosis evaluation of patients with breast cancer.
