目的:采用HPLC/DAD法研究大承气汤的质量控制,同时测定芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、芦荟大黄素、大黄酸、和厚朴酚、厚朴酚、大黄素及大黄酚10种成分的含量。方法:采用AgilentZorbax SB-C18色谱柱(250 mm×4.6 mm,5μm),甲醇-0.1%磷酸水溶液梯度洗脱,流速1.0 mL.min-1,进样量10μL,柱温25℃,双波长检测,芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷的检测波长为280 nm,芦荟大黄素、大黄酸、和厚朴酚、厚朴酚、大黄素、大黄酚的检测波长为254 nm。结果:在文中所述进样量范围内,10种成分进样量与峰面积均呈良好的线性关系,回收率为97.33%~99.95%;RSD为1.32%~2.94%。结论:此方法简单快速灵敏,对于大承气汤的质量控制有一定的参考价值。
OBJECTIVE: To study the quality control of Dachengqi Decoction by HPLC / DAD method and to determine the contents of rutin, naringin, hesperidin, neohesperidin, aloe emodin, rhein, honokiol, Phenol, emodin and chrysophanol content of 10 components. METHODS: The Agilent Zorbax SB-C18 column (250 mm × 4.6 mm, 5 μm) was used to elute gradient of methanol-0.1% phosphoric acid solution with a flow rate of 1.0 mL · min-1. The injection volume was 10 μL and the column temperature was 25 ℃. , Naringin, naringin, hesperidin, neohesperidin detection wavelength of 280 nm, aloe-emodin, rhein, honokiol, magnolol, emodin, chrysophanol detection wavelength of 254 nm. Results: There was a good linear relationship between the injection volume of 10 components and the peak area within the range of injection volume, the recoveries were 97.33% -99.95% and the RSDs were 1.32% -2.94%. Conclusion: This method is simple, rapid and sensitive, and has certain reference value for the quality control of Dachengqi Decoction.
