目的 :探讨 P38信号通道 (P38MAPK)在脂多糖 (L PS)诱导人单核细胞表达肿瘤坏死因子 (TNFα)中的作用。方法 :采用免疫沉淀法、免疫复合物蛋白激酶测定法、Western blotting检测 P38MAPK在 L PS诱导的人单核细胞产生 TNFα中的激活程度 ,应用逆转录聚合酶链式反应 (RT PCR)和酶联免疫吸附法(EL ISA)观察 P38信号通道抑制剂 SB2 0 35 80对单核细胞 TNFα m RNA和蛋白质表达的影响。结果 :L PS刺激单核细胞引起 P38MAPK激活和 TNFα表达明显增多 ,P38MAPK抑制剂 SB2 0 35 80对单核细胞 TNFα表达有显著抑制作用。结论 :P38MAPK可能是 L PS诱导人单核细胞表达 TNFα的重要通路
AIM: To investigate the role of P38 MAPK in the expression of tumor necrosis factor (TNF) induced by lipopolysaccharide (LPS) in human monocytes. Methods: The activation of P38MAPK in LPS-induced human monocyte-derived TNFα was detected by immunoprecipitation, protein kinase immunoassay and Western blotting. Reverse transcription-polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay Immunocytochemistry (ELISA) was used to observe the effect of P38 signal pathway inhibitor SB2 0 35 80 on TNFα mRNA and protein expression in monocytes. Results: LPS stimulated monocytes stimulated P38MAPK activation and TNFα expression was significantly increased, P38MAPK inhibitor SB2 0 35 80 monocytes TNFα expression was significantly inhibited. Conclusion: P38MAPK may be an important pathway of LPS-induced TNFα expression in human monocytes
