目的:探讨缺氧诱导因子 -1α(HIF-1α)和环氧合酶 -2(COX-2)在视网膜新生血管中的表达和意义。方法:以高浓度氧诱导 C57BL/6J 小鼠建立视网膜新生血管模型。取对照组和给氧组幼鼠眼球作荧光素血管灌注,病理切片及免疫组织化学检测,分别观察视网膜血管的改变,视网膜新生血管内皮细胞数目及 HIF - 1α、COX-2 蛋白的表达。结果:给氧组视网膜可见大量新生血管形成,新生血管内皮细胞核数为(23.38±1.07)个,与对照组相比差异有极显著性(P<0.001)。HIF-1α蛋白表达在神经节细胞层和突破视网膜内界膜的新生血管。COX-2 蛋白表达在内核层,神经节细胞层和突破视网膜内界膜的新生血管。两者表达呈显著正相关。(r=0.363,P<0.01)。结论:在视网膜新生血管形成中存在 HIF - 1α、COX-2的高表达,且两者表达密切相关。
Objective: To investigate the expression and significance of HIF-1α and COX-2 in retinal neovascularization. Methods: C57BL / 6J mice were induced to establish retinal neovascularization model with high concentration of oxygen. Fluorescein perfusion, pathological sections and immunohistochemistry were performed in the eyeballs of the control group and the oxygenated group. The retinal vascular changes, the number of retinal neovascular endothelial cells and the expressions of HIF - 1α and COX - 2 protein were observed. Results: A large number of neovascularization was observed in retina of oxygen-fed group. The number of neovascular endothelial cells was (23.38 ± 1.07), which was significantly different from the control group (P <0.001). HIF-1α protein is expressed in the ganglion cell layer and breaks through the neovascularization of the inner limiting membrane of the retina. COX-2 protein is expressed in the nucleus, ganglion cells and the neovascularization that breaks through the inner limiting membrane of the retina. There was a significant positive correlation between the two expressions. (r = 0.363, P <0.01). Conclusion: There are high expression of HIF - 1α and COX - 2 in retinal neovascularization, and the expressions of both are closely related.
