目的:选育紫花丹参多倍体新种质。方法:利用秋水仙碱处理紫花丹参叶片进行多倍体诱导,对其多倍体后代相关性状进行分析。结果:用15 mg/L秋水仙碱浸泡叶片12 h预培养7 d的处理具有较好地加倍效果,加倍率达31.67%。对筛选得到的5种多倍体植株的叶面积、叶绿素含量和主要有效成分含量进行分析表明,多倍体植株较二倍体都有较明显的增加,其中尤以Y2最具应用潜力。结论:通过秋水仙碱诱导获得了紫花丹参多倍体,部分多倍体植株性状变化明显,主要有效成分含量明显提高。
OBJECTIVE: To screen new polyploid germplasm of Salvia miltiorrhiza. Methods: The colchicine of Salvia miltiorrhiza for polyploid induction was used to analyze the related traits of polyploid offspring. Results: Pretreatment of leaves with 15 mg / L colchicine for 12 d pre-culture for 7 days had the best doubling effect with a doubling rate of 31.67%. The analysis of leaf area, chlorophyll content and main active components in the five kinds of polyploid plants showed that polyploid plants had obvious increase compared with diploid, especially Y2 had the most potential application. Conclusion: The polyploid of Salvia miltiorrhiza Bunge was induced by colchicine, and some polyploid plants had obvious changes in the traits. The content of the main active components increased obviously.
