目的:建立白屈菜药材的UPLC-UV特征图谱并用QTOF-MS技术分析其生物碱类成分。方法:采用ACQUITY UPLC BEH C18(2.1mm×100mm,1.7μm)色谱柱,以乙腈-10mmol.L-1醋酸铵缓冲液(用醋酸调节至pH3.0)为流动相梯度洗脱,流速0.4mL.min-1,检测波长280nm,柱温35℃,对10批白屈菜药材的特征图谱进行比较研究,用UPLC-ESI-QTOF-MS技术对其共有峰进行分析。结果:获得白屈菜药材的UPLC特征图谱,共有峰17个,鉴定出14个生物碱成分,其中6个通过对照品对照确认,8个通过质谱信息推测。结论:建立的UPLC-UV特征图谱可用于白屈菜质量评价。
OBJECTIVE: To establish a UPLC-UV profile of Celandine herb and analyze its alkaloid components by QTOF-MS. METHODS: The mobile phase was eluted with ACQUITY UPLC BEH C18 (2.1 mm × 100 mm, 1.7 μm) with acetonitrile-10 mmol.L-1 ammonium acetate buffer (adjusted to pH 3.0 with acetic acid) as mobile phase with a flow rate of 0.4 mL .min-1, the detection wavelength of 280nm, the column temperature of 35 ℃, 10 batches of celandine medicinal characteristics comparison study, using UPLC-ESI-QTOF-MS technology to analyze its common peak. Results: The UPLC characteristic map of celandine herb was obtained. There were 17 peaks in total and 14 alkaloids were identified. Among them, 6 were confirmed by control and 8 were speculated by mass spectrometry. Conclusion: The established UPLC-UV characteristic map can be used to evaluate the quality of Celandine.
