目的探讨兔VX2肿瘤超声造影定量新生血管血流灌注参数与肿瘤组织周细胞变化的关系。方法选用VX2肿瘤肌肉模型兔10只,应用超声造影连续脉冲对比(comparative pulse serial,CPS)技术及ACQ定量分析软件测定肿瘤血管血流灌注各参数值和峰值强度比率(rate of peak index,RPI),并与肿瘤组织标记周细胞的免疫组化染色检测的微血管密度(micro vascular density,MVD)和周细胞数(pericytes number,PN)对照,进行相关性分析。结果肿瘤组织造影血流灌注参数峰值强度(peak intensity,PI)为(15.29±4.14)dB,RPI为(2.95±1.26)dB/s;肿瘤组织标记周细胞的MVD为(17.75±7.60),PN为(26.10±10.59);肿瘤组织造影血流灌注参数PI与MVD和PN显著正相关(r值分别为0.866、0.857,P<0.01),RPI与MVD和PN显著正相关(r值分别为0.647、0.648,P<0.01),而到达时间(arrivaltime,AT)、达峰时间(time-to-peak,TTP)与MVD、PN无相关性。结论超声造影血流灌注参数PI和RPI可以间接反映肿瘤血管生成中周细胞的数量、分布、密度以及微血管密度等变化。
Objective To investigate the relationship between the quantification of perfusion parameters of neovascular VX2 tumor and the change of pericytes in tumor tissue by rabbit VX2 tumor contrast-enhanced ultrasound. Methods Totally 10 VX2 tumor model rabbits were selected. The parameters of vascular perfusion and rate of peak index (RPI) were determined by comparative pulse serial (CPS) and ACQ quantitative analysis software. The correlations were analyzed with the microvascular density (MVD) and pericytes number (PN) detected by immunohistochemical staining of tumor tissue labeled pericytes. Results The peak intensity (PI) of perfusion parameters of tumor tissue was (15.29 ± 4.14) dB and RPI was (2.95 ± 1.26) dB / s respectively. The MVD of pericytes marked by tumor tissue was (17.75 ± 7.60) (26.10 ± 10.59). There was a significant positive correlation between PI and MVD and PN (r = 0.866,0.857, P <0.01), and RPI was positively correlated with MVD and PN (r = 0.647 , 0.648, P <0.01). There was no correlation between arrival time (AT), time-to-peak (TTP) and MVD, PN. Conclusion Contrast-enhanced ultrasound perfusion parameters PI and RPI can indirectly reflect the number, distribution, density and microvessel density of pericytes in tumor angiogenesis.
