目的:应用HPLC建立夏桑菊颗粒的特征图谱,研究配伍对夏桑菊颗粒中夏枯草、桑叶和野菊花特征图谱的影响。方法:Kromasil C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-1.0%冰乙酸溶液(B),梯度洗脱,流速为0.8 m L/min;检测波长为320 nm;柱温为35℃。结果:确定夏桑菊颗粒中的14个特征峰,且对特征峰进行了相应归属:其中6个来源于夏枯草,5个来源于野菊花,3个共同来源于桑叶和野菊花;同时利用对照品指认了夏桑菊颗粒中绿原酸、异迷迭香酸苷、迷迭香酸、蒙花苷4个成分,明确了配伍对这4个成分有较为显著的影响。结论:该方法稳定可靠,可用于夏桑菊颗粒中各单味药夏枯草、桑叶和野菊花特征成分在配伍后变化规律的研究。
OBJECTIVE: To establish the characteristic map of Xiasangju granules by HPLC and study the effect of compatibility on the characteristic patterns of Prunella vulgaris, Morus alba and Chrysanthemum indicum in Xiasangju granules. METHODS: Kromasil C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile (A) -1.0% glacial acetic acid (B) with gradient elution at a flow rate of 0.8 m L / min. The column temperature was 35 ℃. Results: Fourteen characteristic peaks of Xiasangju granules were identified and their corresponding peaks were assigned. Among them, six were from Prunella vulgaris, five from wild chrysanthemum, three from mulberry leaves and wild chrysanthemums. Products identified Xia Seng Ju granules chlorogenic acid, different rosemary acid, rosmarinic acid, monoglyceride 4 ingredients, a clear compatibility of these four ingredients have a more significant impact. Conclusion: The method is stable and reliable, and can be used for the study of the variation of the characteristic components of Prunella vulgaris, Mulberry leaf and Chrysanthemum indicum L. in the single herb medicines of Xia Sang Ju granules.
