淫藤骨痹康方有效部位群在正常大鼠与急性血瘀大鼠体内药物化学对比研究

目的:研究淫藤骨痹康方有效部位群在正常大鼠与急性血瘀大鼠体内药物化学差异性,阐明其活血作用的药效物质基础。方法:采用HPLC法建立淫藤骨痹康方有效部位群在正常生理状态与急性血瘀病理状态下的药物化学特征指纹图谱,并对体内外色谱峰进行对比分析。结果:在正常生理状态下,5个批次的含药血浆样品中有14个共有色谱峰,其中3个峰来自于空白血浆,9个峰为淫藤骨痹康方的原型成分,2个峰为其代谢产物峰或产生的新成分峰,经对照品对照,12 min处的峰为阿魏酸;在急性血瘀病理状态下,5个批次的含药血浆样品中有14个共有色谱峰,其中3个峰来自于空白血浆,9个峰为淫藤骨痹康方的原型成分,2个峰为其代谢产物峰或产生的新成分峰,经对照品对照,12 min处的峰为阿魏酸,32 min处的峰为淫羊藿苷;病理和正常生理状态下扣除空白血浆峰后的共有峰有10个,大多数共有峰峰面积病理状态下明显强于正常生理状态下,其中有3个共有峰峰面积具有显著性差异(P<0.05)。结论:血瘀影响淫藤骨痹康方有效部位群中大多数成分的吸收与代谢;其入血原型成分与代谢新成分可能为其活血作用的药效物质。 OBJECTIVE: To study the pharmacochemical differences between the active part of Kanenji Gubi Kangfang in normal rats and acute blood stasis rats, and to elucidate the pharmacological basis of its active components. Methods: HPLC fingerprint was used to establish the fingerprints of the medicinal features of the active part of Jueguogu Bi-Kang-Fang in normal physiological condition and acute blood stasis. The chromatographic peaks in vivo and in vitro were compared and analyzed. RESULTS: Under normal physiological conditions, there were 14 common chromatographic peaks in 5 batches of drug-containing plasma samples, of which 3 peaks were from blank plasma, 9 peaks were prototype components of JING Teng Gu Bi Bi Kang Fang, 2 Peak for the peak of its metabolites or new peaks produced by the reference substance control, 12 min at the peak of ferulic acid; in the pathological state of acute blood stasis, drug batches of five batches of 14 shared Among them, three peaks were derived from blank plasma, nine peaks were the prototype components of Jueguobubi Kangfang, and two peaks were peaks of their metabolites or new component peaks. The peak was ferulic acid, and the peak at 32 min was icariin. There were 10 common peaks after deducting the blank plasma peak under the pathological and normal physiological conditions, and most of the common peak areas were significantly stronger than the normal physiological state Among them, there are three common peak area with significant difference (P <0.05). CONCLUSION: Blood stasis affects the absorption and metabolism of most components in the active part of Juedeng Gubi Kang Formula, and its prototypical components and new metabolites may be the effective substances for promoting blood circulation.
