目的观察霞草苷对人结肠癌HT-29细胞凋亡的影响及其可能机制。方法用MTT法和流式细胞技术检测浓度为5,10,20,40μmol·L-1霞草苷对HT-29细胞生长、凋亡的影响;用实时荧光定量RT-PCR检测细胞Caspase-3、Bax的mRNA水平。结果霞草苷对HT-29细胞的生长有明显的增殖抑制作用,呈浓度和时间依赖性,细胞凋亡率明显增高。Caspase-3和Bax的mRNA表达量逐渐增加,呈明显的浓度依赖性。结论霞草苷能诱导体外培养的人结肠癌HT-29细胞凋亡,上调Bax表达,活化Caspase-3可能是霞草苷诱导HT-29细胞凋亡的机制之一。
Aim To observe the effect of Xiaoy glycoside on the apoptosis of human colon cancer HT-29 cells and its possible mechanism. Methods The effects of 5, 10, 20, 40μmol·L-Xia Xiaoside on the growth and apoptosis of HT-29 cells were detected by MTT assay and flow cytometry. Caspase-3 , Bax mRNA levels. Results Xia Xiaoside obvious inhibition of proliferation of HT-29 cells in a concentration-dependent and time-dependent manner, the apoptosis rate was significantly increased. Caspase-3 and Bax mRNA expression increased gradually, in a significant concentration-dependent manner. Conclusions Xiaoshuang glycosides can induce the apoptosis of human colon cancer HT-29 cells in vitro, and up-regulate the expression of Bax. Activation of Caspase-3 may be one of the mechanisms that Xiaxiaoside induces HT-29 cell apoptosis.
