目的:探讨白细胞介素1β(IL1β)刺激体外培养的大鼠脑微血管内皮细胞(rCMEC)15羟基前列腺素脱氢酶(15PGDH)活性的经时变化。方法:建立rCMEC培养方法,进行Ⅷ因子相关抗原鉴定。待细胞长至融合状态后,以30ng/ml浓度的IL1β分别刺激0.5、1、2、4、8、12、24h,以放射免疫法测定各刺激时间点15PGDH活性。结果:90%以上的培养细胞呈阳性,确定为rCMEC。加入IL1β刺激后2h,细胞内15PGDH活性与未刺激组比较有显著性差异(P<0.05);12h时达到最大值;24h时有所降低,但与未刺激组比较仍具显著性差异(P<0.01)。结论:rCMEC经IL1β刺激后,细胞内15PGDH活性经历一个逐渐升高,到达峰值后缓慢下降的过程,IL1β刺激12h内皮细胞15PGDH活性达峰值。
AIM: To investigate the temporal changes of 15PGDH activity induced by interleukin 1β (IL1β) in cultured rat cerebral microvascular endothelial cells (rCMECs). Methods: The rCMEC culture method was established to identify Ⅷ factor related antigen. After the cells grew to confluence, the cells were stimulated with IL1β at a concentration of 30ng / ml for 0.5, 1, 2, 4, 8, 12 and 24 hours, respectively, and the 15PGDH activities were measured by radioimmunoassay. Results: More than 90% of cultured cells were positive and identified as rCMEC. The level of 15PGDH in cells after 2h stimulation with IL1β was significantly higher than that in unstimulated group (P <0.05), reached the peak at 12h, decreased at 24h, but still significantly different from that of unstimulated group (P <0.01). CONCLUSION: After stimulated by IL1β, the activity of 15PGDH in rCMEC undergoes a gradual increase and reaches its peak and then decreases slowly. The peak of 15PGDH activity in endothelial cells after IL1β stimulation for 12h is reached.
