应用荧光AFLP和拮抗实验对26株灵芝菌种进行了遗传多样性分析。荧光AFLP分析的结果表明,采用9对EcoR IMse I引物(其中Mse I引物为FAM荧光标记物)对26个菌株进行AFLP扩增,共得到1 944条清晰易辨的多态性条带。聚类分析表明,26株灵芝菌株在遗传相似系数0.28处聚为3类,分别是美国灵芝类、赤芝类和树舌类。种间的遗传相似系数变化范围在0.23~0.73。荧光AFLP分析结果与拮抗性实验结果基本一致,可见拮抗试验在灵芝亲缘关系的初步鉴定中是有效的,荧光AFLP则更能区分出亲缘关系较近的灵芝菌株。
The genetic diversity of 26 Ganoderma lucidum strains was analyzed using fluorescence AFLP and antagonistic experiments. Fluorescent AFLP analysis showed that 9 pairs of EcoR IMse I primers (Mse I primers were FAM fluorescent markers) were used to amplify 26 strains of AFLP, and 1 944 bands were clearly identified . Cluster analysis showed that 26 strains of Ganoderma lucidum clustered into 3 types at genetic similarity coefficient of 0.28, which were American Ganoderma lucidum, Ganoderma lucidum and tree tongue. The genetic similarity coefficient between species ranged from 0.23 to 0.73. Fluorescence AFLP analysis results and antagonistic experiment results are basically the same, it can be seen antagonistic test in the initial identification of genetic relationship Ganoderma lucidum is valid, fluorescent AFLP is more able to distinguish between close Ganoderma lucidum strains.
