目的:观察黑升麻提取物对去势雌性大鼠强迫游泳行为学的影响,探讨黑升麻提取物的抗抑郁效应及安全性。方法3:0只雌性SD大鼠去势后随即分成去势(OVX)对照组、黑升麻(CR)提取物组和美雌醇(E)组,以雌激素药物美雌醇作对照,观察去势雌性大鼠强迫游泳的不动时间及子宫内膜的变化,放射免疫法测定血浆雌二醇含量,采用免疫组织化学的方法,检测下丘脑室旁核c-fos蛋白的表达。结果:与对照组相比,CR、E均可明显缩短强迫游泳大鼠的不动时间(P<0.01),降低下丘脑室旁核神经元中c-fos的表达(P<0.01);但是E组大鼠血浆雌二醇水平明显升高(P<0.01),子宫内膜显著增生;而CR对去势大鼠血浆雌二醇水平无显著影响(P>0.05),子宫内膜无明显改变。结论:CR可以减少强迫游泳大鼠的不动时间,抑制大鼠的抑郁绝望行为,其抗抑郁效应安全有效。
Objective: To observe the effect of black cohosh extract on forced swimming behavior in ovariectomized female rats and explore the antidepressant effect and safety of black cohosh extract. Method 3: 0 female SD rats were divided into ovariectomized (OVX) control group, black cohosh (CR) extract group and estramustine (E) group. The forced swimming time and endometrial changes in ovariectomized female rats were measured. The content of plasma estradiol was determined by radioimmunoassay. The expression of c-fos protein in paraventricular nucleus of hypothalamus was detected by immunohistochemistry. Results: Compared with the control group, CR and E significantly reduced the immobility time in forced swimming rats (P <0.01) and decreased the expression of c-fos in paraventricular nucleus of hypothalamus (P <0.01); however, The plasma estradiol level was significantly increased in E group (P <0.01), and the endometrium was significantly hyperplastic. CR had no significant effect on the plasma estradiol level in ovariectomized rats (P> 0.05) change. Conclusion: CR can reduce forced swimming rats immobility time, inhibit depression and despair in rats, its antidepressant effect is safe and effective.
