目的:探讨人类肺癌细胞株中,不同剂量的多西他赛对真核翻译起始因子3a(eukaryotic in-itiation factor 3 subunit a,eIF3a)和α-微管蛋白(α-tubulin)表达的影响及其时效、量效关系。方法:人类肺腺癌细胞株A549用不同浓度的多西他赛处理不同的时间后,Real-time PCR法检测α-微管蛋白和eIF3a的mRNA表达水平,Western印迹法检测α-微管蛋白和eIF3a的蛋白表达水平。结果:多西他赛不能影响α-微管蛋白mRNA和蛋白水平的表达,且eIF3a与α-微管蛋白的表达水平之间没有明显的相关性。高浓度(30μg/L)多西他赛处理细胞后,eIF3a的mRNA表达水平随着时间的延长有上升的趋势。结论:多西他赛在高剂量时具有增加eIF3amRNA表达的趋势,eIF3a不参与α-微管蛋白表达的调节。
AIM: To investigate the effects of different doses of docetaxel on the expression of eukaryotic in-itiation factor 3 subunit a (eIF3a) and α-tubulin in human lung cancer cell lines And its aging, dose-effect relationship. Methods: Human lung adenocarcinoma cell line A549 was treated with different concentrations of docetaxel for different time. Real-time PCR was used to detect mRNA expression of α-tubulin and eIF3a. Western blotting was used to detect α-tubulin And eIF3a protein expression levels. RESULTS: Docetaxel did not affect α-tubulin mRNA and protein expression, and there was no significant correlation between eIF3a and α-tubulin expression. After treated with docetaxel in high concentration (30μg / L), the mRNA expression of eIF3a tended to increase with time. Conclusion: Docetaxel has the tendency to increase the expression of eIF3amRNA at high dose, and eIF3a is not involved in the regulation of α-tubulin expression.
