[Objective]To detect the contents of β-sitosterol in the roots and stems of Moghani glutiaosa (Prain) Y. T. Wei and M. latifolia Benth. by RP-HPLC-ELSD,as well as the roots of M. philippinensis (Merr. et Rolfe) Li and M. macrophylla (Willd.) O. Kuntze in different grown areas. [Methods] Chromatographic column was Kromasil C18 (250 mm × 4.6 mm,5 μm) ; column temperature was 25 ℃; flow rate was 1. 0 mL/min; mobile phase was methanol. [Results] β-sitosterol showed good linear relationship between 0. 000 65 and 0. 003 25 μg/mL, the average recovery rate was 99. 4% (n = 5) and RSD was 0. 87% . β-sitosterol contents in M. glutiaosa and M. latifolia roots and stems were both relatively high. M. philippinensis grown in Yichang City of Hubei Province had the highest content of β-sitosterol; while M. macro-phylla grown in Huaxi Botanical Garden had the highest content of β-sitosterol. β-sitosterol contents in other grown areas were relatively low. β-sitosterol content in the roots of M. macrophylla was higher than that in M. philippinensis. [Conclusions]This method was simple,rapid and accurate and could be used for the quality control of RADIX MOGHANIAE.
