目的探讨乳腺癌细胞中ERɑ蛋白与p73ɑ蛋白之间物理上及功能上的相互关系。方法采用蛋白印迹法检测ERɑ与p73ɑ的蛋白表达;用免疫共沉淀法检测ERɑ蛋白与p73ɑ蛋白之间的相互结合;用荧光素酶分析实验进一步检测ERɑ与p73ɑ基因的转录活性,以探讨ERɑ与p73ɑ蛋白表达改变的原因。结果 (1)经蛋白酶抑制剂MG132处理细胞后,ERɑ蛋白及p73ɑ蛋白能结合成复合体。(2)共转染ERɑ与p73ɑ时,ERɑ及p73ɑ能相互抑制彼此蛋白表达。结论乳腺癌细胞中ERɑ及p73ɑ能结合形成复合体并能相互抑制彼此蛋白的表达。
Objective To investigate the physical and functional relationship between ERβ and p73ɑ protein in breast cancer cells. Methods The protein expressions of ERɑ and p73ɑ were detected by Western blotting. The co-localization of ERɑ and p73ɑ protein was detected by co-immunoprecipitation. The transcriptional activities of ERɑ and p73ɑ were detected by luciferase assay, The reason for the change of p73ɑ protein expression. Results (1) After the cells were treated with the protease inhibitor MG132, ERɑ protein and p73ɑ protein could be combined into complex. (2) When ERɑ and p73ɑ were co-transfected, ERɑ and p73ɑ could mutually inhibit each other’s protein expression. Conclusion ER ɑ and p73 乳 can combine to form a complex in breast cancer cells and inhibit each other’s protein expression.
