目的 探讨IL - 8在中性粒细胞浸润引起的肝脏缺血再灌注损伤中的作用。方法 建立兔肝缺血再灌注的动物模型 ,于缺血再灌注过程中不同时间点采外周血 ,ELISA法测定IL - 8的浓度 ,同时于相应时间点取肝脏组织作普通病理切片 ,观察中性粒细胞浸润情况 ,并对肝脏病理切片组织作IL - 8免疫组化分析。结果 在肝缺血再灌注肝脏组织中有中性粒细胞浸润和聚集 ,同时随着肝脏组织的破坏 ,引起肝功能损害 ,且在中性粒细胞浸润早期IL - 8免疫组化染色呈阳性反应。结论 在肝脏缺血再灌注过程中 ,由于肝脏组织中IL - 8对中性粒细胞的趋化作用 ,致中性粒细胞在肝脏实质内浸润 ,引起肝细胞破坏 ,释放出各种血清酶 ,从而导致肝脏功能的障碍。这一机制可能是肝缺血再灌注损伤的重要原因
Objective To investigate the role of interleukin-8 (IL-8) in hepatic ischemia-reperfusion injury induced by neutrophil infiltration. Methods The animal model of rabbit hepatic ischemia - reperfusion was established. Peripheral blood was collected at different time points during ischemia - reperfusion. The concentration of IL - 8 was measured by ELISA. At the same time, the liver tissues were taken for routine pathological examination. The infiltration of granulocytes was observed. Immunohistochemical analysis of IL - 8 in liver tissue sections was performed. Results There was neutrophil infiltration and aggregation in liver tissue during liver ischemia - reperfusion, and at the same time with the destruction of liver tissue, liver function was impaired, and IL - 8 immunohistochemical staining was positive in the early stage of neutrophil infiltration . Conclusion During the process of hepatic ischemia - reperfusion, neutrophils infiltrated into the liver due to the chemotactic effect of IL - 8 on neutrophils in the liver, causing the destruction of hepatocytes and releasing various kinds of serum enzymes. Resulting in a malfunction of liver function. This mechanism may be an important cause of liver ischemia-reperfusion injury
