目的:探讨茉莉根醇浸膏对大鼠福尔马林致痛实验的镇痛作用及其可能机制。方法:采用70%乙醇室温浸泡制备茉莉根醇浸膏;采用福尔马林实验观察大鼠Ⅰ相与Ⅱ相的疼痛反应;用紫外分光光度法测定福尔马林实验大鼠血清中PGE2含量;Elisa法测定皮层、丘脑和海马中5-羟色胺(5-HT)、多巴胺(DA)、去甲肾上腺素(NE)含量来研究药物的可能镇痛机制。结果:茉莉根醇浸膏能明显减轻福尔马林致痛大鼠Ⅰ相与Ⅱ相的疼痛反应;茉莉根醇浸膏不能降低福尔马林实验大鼠血清中PGE2的含量,但能明显升高大鼠皮层和丘脑中5-HT含量,也能明显升高皮层、丘脑和海马中DA和NE的含量;结论:茉莉根醇浸膏对大鼠福尔马林致痛实验具有镇痛作用,其镇痛机制与外周炎性致痛因子PGE2无关,与升高皮层和丘脑中5-HT含量,以及升高皮层、丘脑与海马中DA和NE含量有关。
Objective: To investigate the analgesic effect of jasmine root alcohol extract on rat induced by formalin and its possible mechanism. Methods: The jasmine root alcohol extract was prepared by soaking 70% ethanol at room temperature. The pain reaction of phase Ⅰ and phase Ⅱ in rats was observed by formalin test. The content of PGE2 in serum of formalin test rats was determined by ultraviolet spectrophotometry Elisa method was used to study the possible analgesic mechanism of drugs by measuring the content of 5-HT, DA and NE in cortex, thalamus and hippocampus. Results: Jasmine root alcohol extract can significantly reduce the pain reaction of phase Ⅰ and phase Ⅱ in formalin-induced rat pain. Jasmine root alcohol extract can not reduce the content of PGE2 in serum of formalin test rats, Elevated 5-HT level in rat cortex and thalamus also significantly increased the content of DA and NE in cortex, thalamus and hippocampus. Conclusion: Jasmine root alcohol extract has analgesic effect on rat induced by formalin , And its analgesic mechanism has nothing to do with the peripheral inflammatory pain factor PGE2, which is related to the increase of 5-HT content in cortex and thalamus, as well as the increase of DA and NE in cortex, thalamus and hippocampus.
