目的:对江南卷柏药材进行生药鉴定研究,以制定其药材的质量标准。方法:观察江南卷柏药材的性状和显微鉴别特征;以穗花杉双黄酮为对照品进行TLC色谱研究;采用HPLC法建立江南卷柏药材的含量测定方法。结果:TLC法采用硅胶GF254板,以甲苯-甲酸乙酯-甲酸(5∶4∶0.5)为展开剂,FeCl3乙醇溶液为显色剂,可出现4个清晰的斑点;HPLC法以穗花杉双黄酮为对照品,以乙腈-水(含3%四氢呋喃和3%三氟醋酸)为流动相,检测波长330 nm,经对3批药材测定,药材中穗花杉双黄酮含量不少于0.4%。结论:江南卷柏药材的显微、TLC鉴别和穗花杉双黄酮的含量测定等方法准确可靠,可有效控制江南卷柏药材的质量。
OBJECTIVE: To study the crude drug identification of southern Chinese cypress medicinal materials to establish its quality standard. Methods: Observe the properties and microscopic identification characteristics of the medicinal materials of Selaginella concinnaceae. TLC chromatogram was carried out by using the biflavonoids of Radix Aconiti kusnea as reference substance. The HPLC method was used to determine the content of Selaginella concinna. Results: TLC method using silica gel GF254 plate, with toluene - ethyl formate - formic acid (5: 4: 0.5) as developing agent, FeCl3 ethanol solution as developer, there are 4 clear spots; Flavonoids were used as reference substance. The mobile phase consisted of acetonitrile-water (containing 3% tetrahydrofuran and 3% trifluoroacetic acid) as the mobile phase. The detection wavelength was 330 nm. Conclusion: The methods of microscopical, TLC identification and determination of the content of biflavonoids in Panax quinquefolius are accurate and reliable, which can effectively control the quality of Selaginella concinna.
